Accuracy of a Novel “Factory-Calibrated” Continuous Glucose Monitoring Device in Normal Glucose Levels: A Pilot Study

Abstract: Free Style Libre Pro, a novel continuous glucose monitoring (CGM) device for retrospective assessment of glycemic excursion, does not require calibration during the measurement period. In this study, the accuracy of FreeStyle Libre Pro in normal glucose levels was investigated. Two meal tests, on the 7th day and 9th day of CGM usage, were performed in five non-diabetic adults. Venous blood samples were obtained at baseline, and at 2 hours, 4 hours and 6 hours after the meal. The plasma glucose levels and the glucose levels measured by FreeStyle Libre Pro were compared. Subjects were aged 41.4±7.6 years, 40% male, with body mass index 21.7±2.6, HbA1c 5.6±0.3% and a fasting plasma glucose level of 95.4±2.1 mg/dl. The plasma glucose levels and the glucose levels measured by FreeStyle Libre Pro were significantly different; 94.4±1.8 vs. 111.2±8.5 mg/dl (P <0.01) at baseline, 95.6±7.0 vs. 122.6±8.8 mg/dl (P <0.01) at 2 hours, 90.1±4.3 vs. 111.2±13.0 mg/dl (P <0.01) at 4 hours, and 88.7±3.2 vs. 105.5±7.1 mg/dl (P <0.01) at 6 hours. The absolute difference between the two methods was 17.7±7.5 (minimum 5 - maximum 27) mg/dl at baseline, 27.0±9.4 (minimum 8 - maximum 40) mg/dl at 2 hours, 21.1±11.3 (minimum 7 - maximum 42) mg/dl at 4 hours, 16.8±5.4 (minimum 7 - maximum 23) mg/dl at 6 hours, and 20.7±9.3 (minimum 5 - maximum 42) mg/dl for all time points, respectively. The absolute relative difference between them was 18.7±7.8 (minimum 5.3 - maximum 28.7)% at baseline, 28.7±10.7 (minimum 7.7 - maximum 45.5)% at 2 hours, 23.3±12.2 (minimum 7.9 - maximum 45.2)% at 4 hours, 18.9±6.0 (minimum 8.0 - maximum 26.2)% at 6 hours, and 22.4±10.0 (minimum 5.3 - maximum 43.5)% for all time points, respectively. The ratio of values measured by FreeStyle Libre Pro within the interval of ±15 mg/dl in less than 100 mg/dl or within the interval of ±15% in not less than 100 mg/dl plasma glucose levels was 30.0% at baseline, 10.0% at 2 hours, 40.0% at 4 hours, 50.0% at 6 hours and 32.5% for all time points, respectively. These study results suggest FreeStyle Libre Pro may not be sufficiently accurate in normal glucose levels.

Abstract: Osteoarthritis (OA) is characterized by the degeneration of affected joints leading to pain and reduced mobility. Periostin appears as a potential candidate in transducing mechanical signals to stimulate chondrocyte proliferation. 15 rats (E-group) underwent both medial meniscectomy and medial collateral ligament (MCL) transaction, while the other 15 rats (S-group) underwent sham surgery. Animals were killed at 1, 2, and 4 weeks postsurgery, and 5 animals were put into use per-time point in each treatment group. Cartilage was harvested from tibial plateau and femoral condyle. We examined the level of Postn expression in cartilage of the experimental OA, using real-time PCR and immunohistochemistry analysis. The expression of Postn was about 2-fold higher at 1 week (P=0.08<0.05), 4-fold higher at 2 weeks (P=0.003<0.05) and 4-fold higher at 4 weeks (P=0.034<0.05) post-surgery in E-group compared to S-group. Postn protein expression levels were markedly increased in the E-Group compared with the S-Group at 1, 2 and 4 weeks post-surgery. This study will be of benefit to those subsequent studies using the OA model to evaluate the outcomes of Postn epxpression in mechanism of OA. Postn may play prominent roles in pathogenic mechanisms of OA.

Abstract: Herbomineral formulations have been used and accepted all around the world owing to its medicinal and therapeutic importance. The aim of the study was to evaluate the immunomodulatory potential of Biofield Energy Healing (The Trivedi Effect®) Treatment on the new proprietary herbomineral formulation in male Sprague Dawley rats. The test formulation was divided into two parts. One part was denoted as the control without any Biofield Energy Treatment, while the other part was defined as the Biofield Energy Treated sample, which received the Biofield Energy Healing Treatment remotely from twenty renowned Biofield Energy Healers. The Biofield Energy Treated and untreated test formulation was evaluated using experimental parameters such as humoral immune response (primary and secondary antibody titre), delayed type hypersensitivity reaction, animal weight parameters, feed and water intake, histopathology, hematological and serum biochemistry were studies. Humoral immune data showed the primary and secondary antibody titre values were significantly (p≤0.05) increased by 105.88% and 260%, respectively in the Biofield Energy Treated Test formulation (G4) group with respect to the disease control group (G2). The delayed type hypersensitivity results showed a significant increased paw edema by 32.43% and 53.33% at 24 and 48 hours, respectively in the Biofield Energy Treated Test formulation (G4) group, while untreated test formulation group (G5) animals showed 13.33% increase in the paw edema (at 48 hour) compared with the G2 group. Hematological analysis showed a significant increase in the red blood cell count, while the platelet count was significantly increased by 27.78% in Biofield Energy Treated Test formulation (G4) group as compared with the G2 group. Biochemical analysis exhibited an increased level of calcium and phosphorus by 1.58% and 12.65%, respectively while serum uric acid was significantly decreased by 36.66% in the G4 group compared with the G2 group. In addition, animal weight parameters like body weight, relative organ weight, histopathology, water and feed intake data suggest that the Biofield Energy Treated herbomineral formulation was found safe, non-toxic, throughout the exposure period. In conclusion, the The Trivedi Effect®-Biofield Energy Healing based herbomineral formulation would be the powerful immunomodulatory product, which can be useful to modulate the immunity against many infectious diseases. Overall, experimental data suggests that the Biofield Energy Treated test formulation can be used for autoimmune and inflammatory diseases, stress management and prevention, and anti-aging by improving overall health.
